چکیده

The inability of the methionine requiring Escherichia coli mutant (1-344) to grow in methionine free media containing inorganic sulfur, cystine, or homocystine has been demonstrated (Lampen et al., 1947). While methionine has been shown to be essential for growth, the metabolism of nonmethionine sulfur added to media containing methionine has not been investigated directly. In the results reported here we have attempted to observe some of the metabolic reactions occurring on either side of the biochemical block, and to determine what degree of coupling exists between the necessary utilization of methionine and the metabolism of inorganic sulfur.

Metabolism of selenium by Escherichia coli: biosynthesis of selenomethionine.

Selenium, an element long known to be toxic to living organisms (1, 2), has only recently been implicated as an essential trace element for rats (3), chicks (4,5) and several other animals. The work of Franke (6), Horn et cd. (7), Painter (8, 9), and McConnell (10, 11) established that if toxic amounts of selenium were supplied in various chemical forms to a number of plants, animals, and micro...

Sulfur Metabolism in Enterobacteria 145 Sulfur Metabolism in Escherichia coli and Related Bacteria: Facts and Fiction

Living organisms are composed of macromolecules made of hydrogen, carbon, nitrogen, oxygen, phosphorus and sulfur. Much work has been devoted to the metabolism of the first five elements, but much remains to be understood about sulfur metabolism. We review here the situation in Escherichia coli and related bacteria, where more than one hundred genes involved in sulfur metabolism have already be...

Conversion of methionine to cysteine in Bacillus subtilis and its regulation.

Bacillus subtilis can use methionine as the sole sulfur source, indicating an efficient conversion of methionine to cysteine. To characterize this pathway, the enzymatic activities of CysK, YrhA and YrhB purified in Escherichia coli were tested. Both CysK and YrhA have an O-acetylserine-thiol-lyase activity, but YrhA was 75-fold less active than CysK. An atypical cystathionine beta-synthase act...

Studies on the Sulfur Metabolism of Tissues Cultivated in Vitro Iv. Ergothioneine, Oxidized Glutathione, and Lanthionine by Helen

Previous studies from this Laboratory (1, 2) have shown that chick embryonic heart fibroblasts cultivated in vitro in a completely synthetic medium (3, 4) have an essential requirement for n-cystine plus a supplementary requirement for Lor n-methionine. Of more than thirty sulfur-containing compounds tested, only L-cysteine was found to replace L-cystine completely (5), although moderate replac...
